Radio HPLC Detectors WHAT IS RADIOCHROMATOGRAPHY? Chromatography is a laboratory method performed to separate the different components of a mixture. The mixture is dissolved in a fluid called the mobile phase. The mobile phase carries the mixture through a column which is the stationary phase. The various compounds of the mixture travel at different speeds through the column causing them to separate. In order to measure the separation of the mixture the compounds must be detectable in some way. There is no universal detector that can monitor all compounds, and therefore different types of HPLC detectors must be used depending on the properties of the substances to be separated. UV detectors are often used, as many substances typically separated in HPLC absorb light in the UV range of light. Other detectors such as, refractive index, light scattering, fluorescence, conductivity, and more, are also used in HPLC. The intensity of the signal measured will be represented in the graph called chromatogram. Assuming complete separation, each compound will appear in the chromatogram as a peak. The time of appearance identifies the compound, and the area of the peak quantifies it. In the case of Radio-HPLC (or radio chromatography), the substances to be separated are radioactive because they have been labeled using a radioactive isotope, such as 125I, 14C or 3H. APPLICATIONS OF RADIO-HPLC Radio-HPLC has numerous applications in research, diagnostics, and quality control. Some of the most common and important applications are: Pharmaceutical Analysis: Radio-HPLC is a key technology for the separation and quantitation of radiolabeled drugs and putative metabolites in drug development. Environmental Analysis: A wide variety of contaminants from sources such as industrial waste, landfill sites, pesticides and pharmaceutical drugs can make their way into the environment. The challenging identification of these contaminants require a highly sensitive detector to be able to detect the lowest concentrations of sample. Development and QC of Radio chemicals: Radiochemicals are only as good as the care taken in each preparation step and strict regulations must be met during production. Radiochemical purity analysis using Radio HPLC is often required to verify the quality of the produced radiochemicals. QC of Radiolabeled Antibodies and Proteins: Radiochemical identity and purity analysis of radiolabeled antibodies and proteins using Radio-HPLC is a simple and reliable method to ensure that only the radiolabeled antibody or protein is present in the quality control sample.

The HERM is a dedicated radio HPLC monitor which can be connected to different detectors providing flexibility, reproducibility and sensitivity. The HERM with NaI detector for gamma and high energy beta isotopes combines an extremely sensitive NaI detector with compact shielding and can detect even low energy gamma isotopes like 125I with high efficiency. Simple integration into every HPLC system

The HERM makes integration into any existing HPLC system very easy. The PC connection can be established instantly via the USB port or by using the analog 0-1 V output. Its compact design makes it easy to fit in to a hot cell.

Sensitive and reliable measurement

The NaI detector option combines an extremely sensitive detector with a compact shielding. Even low energy gamma isotopes like 125I can be detected with high efficiency. Lead shielding ensures low backgrounds even when there is high external radiation.

Easy operation and high flexibility

The HERM LB 500 has been designed to be intuitive and easy to use. This will get you up and running as quickly as possible. Standard tubing is used to create the flow cell volume and provides maximum flexibility. Changing cells is very easy because of the twin cell changer that enables the user to easily switch between two different cell volumes. If required by GLP regulations, the cell volume can be fixed to just one volume using the optional single volume insert. The system has been designed to avoid accidental parameter modifications eliminating manual adjustment as all instrument parameters are controlled remotely.

Dual operation mode

The HERM system can be installed in multiple distinct configurations, providing more flexibility and convenience. The comfortable stand-alone configuration provides an analog signal which can be easily integrated into an existing HPLC system. The system is equipped with a flash memory and a powerful CPU. Alternatively, the system can be installed in the PC-controlled configuration via a USB connection for digital data acquisition.

Easy-to-use software for maximum productivity

The HERM Controller is the software interface to the instrument; it controls all functions and collects the data. The RadioStar software combines the ability to run sophisticated measurements with a user-friendly interface. A Chromeleon™ driver is available, Chromeleon™ is known as a very versatile chromatography software package supporting many different hardware solutions. The driver is fully certified and digitally signed by Thermo Scientific™ and runs with versions 6.x and 7.x.
